Your top floor, and maybe the 2nd floor should allways be 3 wides and 2 wides (don't upgrade all these rooms yet, not untill you have strong enough wpns & dwellers to defend the), for defence versus raiders that break down your vaults front gate and come charging in for loot and shennanigans. Unfortunately, there is no way to move rooms directly.
